ABSTRACT

Parodontidae is a relatively small group of Neotropical characiform fishes consisting of three genera (Apareiodon, Parodon, and Saccodon) with 32 valid species. A vast cytogenetic literature is available on Apareiodon and Parodon, but to date, there is no cytogenetic data about Saccodon, a genus that contains only three species with a trans-Andean distribution. In the present study the karyotype of S. wagneri was described, based on both conventional (Giemsa staining, Ag-NOR, C-bands) and molecular (repetitive DNA mapping by fluorescent in situ hybridization) methods. A diploid chromosome number of 2n = 54 was observed in both sexes, and the presence of heteromorphic sex chromosomes of the ZZ/ZW type was detected. The W chromosome has a terminal heterochromatin band that occupies approximately half of the long arm, being this band approximately half the size of the Z chromosome. The FISH assay showed a synteny of the 18S-rDNA and 5S-rDNA genes in the chromosome pair 14, and the absence of interstitial telomeric sites. Our data reinforce the hypothesis of a conservative karyotype structure in Parodontidae and suggest an ancient origin of the sex chromosomes in the fishes of this family.(AU)


Parodontidae é um grupo relativamente pequeno de peixes caraciformes neotropicais que consiste em três gêneros (Apareiodon, Parodon e Saccodon) com 32 espécies válidas. Uma vasta literatura citogenética está disponível sobre Apareiodon e Parodon, mas até o momento não há dados citogenéticos sobre Saccodon, um gênero que contém apenas três espécies com distribuição transandina. No presente estudo foi descrito o cariótipo de S. wagneri, baseado em métodos convencionais (coloração de Giemsa, Ag-NOR, bandas C) e moleculares (mapeamento de DNA repetitivo por hibridização fluorescente in situ). Um número cromossômico diplóide de 2n = 54 foi observado, e a presença de cromossomos sexuais heteromórficos do tipo ZZ/ZW foi revelada. O cromossomo W possui uma banda terminal heterocromática que ocupa aproximadamente metade do braço longo, sendo esta banda aproximadamente a metade do tamanho do cromossomo Z. O ensaio FISH mostrou uma sintenia dos genes 18S-rDNA e 5S-rDNA no par de cromossomos 14, e a ausência de sítios teloméricos intersticiais. Nossos dados reforçam a hipótese de uma estrutura cariotípica conservadora em Parodontidae e sugerem uma origem ancestral dos cromossomos sexuais nos peixes desta família.(AU)

ABSTRACT

Ameivula is as a new genus of Teiidae family that emerged after extensive revision of species that comprised the former complex of species called Cnemidophorus group. Its species has a wide distribution from the northeast of Brazil to northern Argentina. Cytogenetic studies in the Teiidae family have shown that karyotypical data are important tools in phylogenetic and systematic studies within this group allowing to determine the position of species in the family. Thus, this study aimed to describe the karyotype of Ameivulaocellifera (Spix, 1825) from Picos, Piauí state in the Brazilian Northeast. Specimens were collected from August 2014 to October 2015 using interception traps and pitfalls, mounted randomly along the Caatinga area. The animals were collected and transported to Federal Institute of Piauí, campus Picos, where was carried out all laboratory procedures. Individuals analyzed showed a diploid number of 2n = 50 for both sexes, with karyotype composed by 30 macrochromosomes and 20 microchromosomes of telocentric and subtelocentric types. There were no heteromorphic sex chromosomes in the studied specimens. C-band technique evidenced the heterochromatic blocks in pericentromeric and telomeric regions of chromosomes. The nucleolar organizing regions appeared as a simple unit located at the terminal portion of the long arm of chromosomal pair number 5. The chromosomal characteristics of A. ocellifera analyzed do not show divergences regarding individuals from other regions. However, the nucleolar organizing regions seems to be a good chromosomal marker that permits to distinguish the species already studied.


Ameivula é um novo gênero da família Teiidae que surgiu após extensa revisão de espécies que compuseram o antigo complexo de espécies chamado grupo Cnemidophorus. Suas espécies têm uma ampla distribuição do nordeste do Brasil ao norte da Argentina. Estudos citogenéticos na família Teiidae mostraram que os dados cariotípicos são ferramentas importantes em estudos filogenéticos e sistemáticos dentro deste grupo, permitindo determinar a posição das espécies na família. Assim, este estudo tem como objetivo descrever o cariótipo de Ameivula ocellifera (Spix, 1825) de Picos, no nordeste brasileiro. Os espécimes foram coletados de agosto de 2014 a outubro de 2015 utilizando armadilhas de interceptação e armadilhas, montadas aleatoriamente ao longo da área da Caatinga. Os animais foram coletados e transportados para o Instituto Federal do Piauí, campus Picos, onde foram realizados todos os procedimentos laboratoriais. Os indivíduos analisados apresentaram um número diploide de 2n = 50 para ambos os sexos, com cariótipo composto por 30 macrocromossomos e 20 microcromossomos dos tipos telocêntrico e subtelocêntrico. Não houve cromossomos sexuais heteromórficos nos espécimes estudados. A técnica da banda C evidenciou os blocos heterocromáticos nas regiões pericentroméricas e teloméricas dos cromossomos. As regiões organizadoras de nucléolos apareceram como uma unidade simples localizada na porção terminal do braço longo do par cromossômico número 5. As características cromossômicas de A. ocellifera analisadas não mostram divergências em relação a indivíduos de outras regiões. No entanto, as regiões organizadoras de nucléolos parecem ser um bom marcador cromossômico que permite distinguir as espécies já estudadas.

ABSTRACT

Resumo Estudos mostram que, a despeito da queda das taxas de mortalidade e das melhorias nas condições de saúde, os trabalhadores têm saído mais cedo da atividade econômica. Esperar-se-ia que uma vida mais longa aumentasse os retornos aos investimentos em capital humano. A literatura associa a saída precoce do mercado de trabalho à cobertura da seguridade social. O adiamento da idade à aposentadoria é considerado uma alternativa para conter o desequilíbrio fiscal em muitos países. No entanto, estudos sugerem a existência de barreiras que inibem a permanência do trabalhador mais velho na atividade econômica. Entre elas, cita-se o preconceito por parte dos empregadores. O objetivo deste trabalho é buscar entender a não participação de homens brasileiros de 50-64 anos nas atividades econômicas, aí incluídos os que não estão aposentados (nem-nem). São o primeiro grupo a ser afetado pela reforma previdenciária proposta pelo governo. A proporção desses no total desta faixa etária aumentou de 3,5% para 10,2%, entre 1984 e 2017. Baixa escolaridade e piores condições de saúde em relação aos demais homens podem dificultar essa inserção. Isso sugere uma discriminação com relação aos trabalhadores mais velhos e a falta de políticas públicas que visem reforçar a capacidade destes indivíduos para conseguir um emprego.


Abstract Several studies show that despite a decline in mortality and improvements to health conditions, workers have left the economic activities early. The literature associates precocious exit from the labour market to the widespread coverage of Social Security. One alternative to contain the fiscal imbalance in most countries has been to postpone the minimal age to be entitled to a pension benefit. Nevertheless, many studies suggest the existence of barriers that make it difficult for older workers to remain in economic activity. Among them are prejudices among employers.This paper aims to understand the non-participation of Brazilian men aged 50-64 in economic activities. The focus on this age group is because they would be the first group to be affected by the pension reform proposed by the Government. These are those who are neither in the labour market nor retired (neither-nor). The proportion of these men of the total number of men in this age group increased from 3.5% to 10.2% between 1984 and 2015. Very low schooling and worse health conditions compared to other men can contribute to difficulties for insertion. This suggests discrimination in relation to the older worker and the lack of public policies aimed at reinforcing the ability of these individuals to obtain a job.

ABSTRACT

Background: Diarrhea is a condition of altered intestinal water and electrolyte transport. The pathophysiologic mechanisms of diarrhea include osmotic, secretory, inflammatory, and altered motility. Hence; we planned the present study to assess and compare the efficacy of ofloxacin and combination of Norfloxacin and metronidazole (Nor Metrogyl) in treating patients with acute watery diarrhoea. Materials & Methods: The present study was conducted in the department of general medicine of RBM Hospital, Bharatpur, Rajasthan and included comparative evaluation of efficacy of ofloxacin and combination of Norfloxacin and metronidazole (Nor Metrogyl) in treating patients with acute watery diarrhoea.  A total of 24 patients with chief complaint of acute watery diarrhea were included in the present study and were randomly divided into two study groups; group A and group B. group A included subjects who were treated with ofloxacin therapy while group B included subjects who were treated with Nor Metrogyl. All the results were analyzed by SPSS software. Results: A total of 24 patients were included in the present study and were randomly divided into two study groups with 12 patients in each group. We observed non-significant results while comparing the clinical success and biological success among patients of both the study groups. Conclusion: Both the treatment regimens are equally effective in treating diarrhea patients.

ABSTRACT

Objective: To study triterpenoids of Paliurus ramosissimus. Methods: Six triterpenoids were obtained by modern chromatography technologies, such as repeated chromatography on silica gel, ODS, Sephadex LH-20, and preparative HPLC. Their structures were elucidated on the basis of combination of IR, mass spectrometry, and NMR spectroscopy. Results: Six triterpenoids were isolated from the ethyl acetate soluble parts obtained from 95% aq. ethanol extract of the aerial parts of P. ramosissimus. They were identified as 22β,24-dihydroxy-A(1)-nor-2,20(29)-lupindien-27,28-dicarboxylic acid (1), zizyberenalic acid (2), 3-O-protocatechuoylceanothic acid (3), ceanothetric acid (4), 2-O-caffeoylalphitolic acid (5), and 3-O-caffeoylalphitolic acid (6). Conclusion: Compound 1 paliurus ramosissimus acid A is a new compound. Compounds 2-6 are isolated from P. ramosissimus for the first time. And the complete NMR data of compound 3 has been presented in this paper for the first time.

ABSTRACT

Nor-ursane triterpenes are formed by the reduction of one or several methyl groups on the ursane triterpene carbon skeleton. According to the position of vacant methyl, they can be divided into eight types, including 23-nor, 24-nor, 28-nor, 29-nor, 30-nor, 2-nor, 3-nor, and 3,24-nor. Moreover, nor-ursane triterpenes have several pharmacological effects such as anti-cancer, anti-inflammatory, anti-bacterial, inhibition of acyl cholesterol acyl transferase (ACAT) and so on. In this study, more than 30 relevant literatures of nor-ursane triterpenoid compounds at home and abroad were reviewed. The plant distribution, pharmacological activity and structural characteristics of nor-ursane triterpenes were studied. Therefore, this review may provide the basis for further study of this type of compound.

ABSTRACT

El cannabis es una de las drogas ilegales más usadas a nivel mundial. Su consumo se relaciona con diferentes hechos en el ámbito forense, laboral, deportivo y clínico. Para su detección se utilizan métodos con diferentes fundamentos y alcances (inmunológicos, cromatográficos). En este trabajo se describe un método preciso, reproducible y validado, para la cuantificación del principal metabolito urinario del Δ9-tetrahidrocannabinol, el ácido-11-nor-9-carboxi- Δ 9-tetrahidrocannabinol (THC-COOH), por cromatografía gaseosa-espectrometría de masas (GC-MS). Se efectuó una extracción en fase sólida (SPE) a partir de la orina, previa hidrólisis alcalina. Se utilizó el análogo deuterado (THC-COOH D3) como estándar interno. El análisis por GC-MS se realizó en modo SIM. La curva de calibración fue lineal en el rango de trabajo (10-100 ng/ml, r > 0,999) y el límite de cuantificación fue de 10 ng/ml. La recuperación absoluta estuvo comprendida entre el 91,0 y 99,0 %. La precisión intra e inter ensayo fue de 1,06 a 1,26 y 3,59 a 9,80 %, respectivamente. El método fue aplicado a muestras reales, positivas por test inmunológico, resultando ser muy útil y fiable en el análisis de rutina de THC-COOH en orina humana con fines toxicológicos.


Cannabis is one of the most widely used illegal drug in the world. Its consumption is related to different forensic, work, sports and clinical events. In order to determinate the presence of cannabis, different methods with distinct fundamentals and scopes (immunoassay and chromatography) are applied. This report described an accurate, reproducible, and validated gas chromatography-mass spectrometry (GC-MS) method for the quantitation of 11-nor-9-carboxy- Δ9-tetrahydrocannabinol (THCCOOH), the major metabolite of Δ9-tetrahydrocannabinol in urine. A solid phase extraction (SPE), previous alkaline hydrolysis, was performed on the urine sample. Its deuterated analog (THC-COOH D3) was used as internal standard. The GC-MS analysis made by selected ion monitoring (SIM). Calibration curve was linear over the specified range (10 -100 ng/ml; r > 0.999) and limit of quantitation was 10 ng/ml. Absolute recoveries ranged from 91.0 to 99.0. Intra-assay and inter assay precision ranged from 1.06 to 1.26 and 3.59 to 9.80 %, respectively. The method has been applied to real samples, positive to immunological screening test, resulting to be very useful and reliable in routine analysis of THC-COOH in human urine for toxicological purposes.

ABSTRACT

Background: Duloxetine is relatively considered as a treatment for diabetic neuropathy pain due it is balanced and potent reuptake inhibitor of both serotonin and nor epinephrine where these neurotransmitters play a great in pain inhibition. Materials and Methods: We searched DLX related articles in Pubmed, Cochrane and Embase from 2005 to 2010. 158 articles were found after through search out of which 68 articles were case reports, reviews and meta-analysis, 40 studies were clinical trials but not efficient data was available, 45 studies were RCTs but not related to our topic. Only 5 RCTs included after exclusion. We then performed the meta-analysis of the studies which met our eligibility criteria we performed fixed effect model network meta-analysis to analyze the efficacy of DLX compare to placebo. We chose diabetic neuropathy duration, diabetes mellitus duration, types of diabetes and DLX with regard to MNSI scale. Results: 5 published RCTs were included in this meta-analysis no significant difference observed for DLX and DND [(SD mean difference 0.22 (95%CI -0.16 to 0.60); P=0.25)] , on diabetes mellitus duration for HbA1c and fasting blood glucose (FBG) [(SD mean difference -0.00 (95%CI-0.087 to 0.87); P=1.00)] and on types of diabetes[(COR 1.00 (95% CI 0.73 to 1.38; P=0.98)] and [(OR 1.00 (95% CI 0.72 to 1.37); P=0.98)] .DLX shown to have significant efficacious compared to placebo for MNSI (95% CI -0.37 to – 0.03); P=0.02)]. Conclusion: In the fixed effect model analyses of DLX, showed similar efficacy to placebo for efficacy parameters, except on MNSI scale but not clinically relevant.

ABSTRACT

The phytochemistry investigation on the Cassia occidentalis, a Dai Medicine, was carried out. The C. occidentalis was extracted with ethanol and then partitioned with EtOAc. The EtOAc soluble materials were subjected repeatedly to column chromatography on silica gel and preparative RP-HPLC, leading to isolation of a nor-sesquiterpene, 3-isopropyl-1,6-dimethoxy-5-methyl-naphthalen-7-ol (1), and a sesquiterpene, 2,7-dihydroxy-4-isopropyl-6-methyl-naphthalene-1-carbaldehyde (2). Their structures were determined by means of spectroscopic studies. Compound 1 is a new compound. Compound 2 is also isolated from C. occidentalis for the first time. In addition, the cytotoxicity of compound 1 for NB4, A549, SHSY5Y, PC3, and MCF7 cells line was assayed by using the MTT method, and it displayed potential cytotoxicity for the tested cancer cell-line with IC₅₀ valves of (1.8±0.2), (1.2±0.2), (0.9±0.1), (2.2±0.3), (2.6±0.3) μmol•L⁻¹, respectively.

ABSTRACT

Hypoptopomatinae is a monophyletic subfamily that includes 147 species, distributed in 20 genera. Otothyropsis is a genus of Hypoptopomatinae, recently described. Here, we provided the first cytogenetic information of Otothyropsis . The specimens were collected from córrego Dourado, a small tributary of rio Iguatemi, which flows into rio Paraná. The specimens of Otothyropsis cf. polyodon were analyzed with respect to diploid number, C-Band and Ag-NOR patterns. The diploid number was 54 chromosomes, distributed in 18 metacentric, 28 submetacentric, and 8 subtelocentric chromosomes, with single Ag-NOR and conspicuous heterochromatic blocks on the short and long arms of the 24th pair of chromosomes. Our study highlights the conservation trend of the diploid number (2n=54) and fundamental number (FN = 108) among the species of Hypoptopomatinae. However, the karyotype formula (18m+28sm+8st) seems to be specific to O. cf. polyodon , considering the other Hypoptopomatinae species already analyzed.


Hypoptopomatinae é uma subfamília monofilética que inclui 147 espécies distribuídas em 20 gêneros, sendo Otothyropsis um gênero recentemente descrito. Aqui, fornecemos a primeira informação citogenética do gênero Otothyropsis . Espécimes foram coletados no córrego Dourado, um pequeno tributário do rio Iguatemi, o qual deságua no rio Paraná. Espécimes de Otothyropsis cf. polyodon foram analisados em relação ao número diploide e padrões de Banda-C e Ag-NOR. O número diploide foi de 54 cromossomos, distribuídos em 18 metacêntricos, 28 submetacêntricos e 8 subtelocêntricos, com Ag-NOR simples e blocos heterocromáticos evidentes no braços curto e longo do par de cromossomos 24. Nosso estudo destaca a tendência de conservação do número diploide (2n=54) e número fundamental (NF=108) entre as espécies de Hypoptopomatinae. Entretanto, a fórmula cariotípica (18m+28sm+8st) parece ser específica para O. cf. polyodon, considerando as outras espécies de Hypoptopomatinae já analisadas.

ABSTRACT

A coloração pela prata das regiões organizadoras de nucléolos (NORs) é caracterizada por marcar proteínas ligadas ao ácido ribonucleico ribossômico, avaliando a proliferação em células normais ou neoplásicas. Objetivou-se estudar, em testículos de ovinos obtidos em matadouro, a validade do uso da técnica de coloração pela prata (AgNOR) na identificação das regiões organizadoras de nucléolo (NORs) em células saudáveis da linhagem espermatogênica. Utilizaram-se 43 pares de testículos de ovinos mestiços entre seis e 10 meses de idade. Testes de Wilcoxon e Spearman foram empregados, com nível de 5%. As médias das NORs nas células das gônadas direita e esquerda foram, respectivamente: espermatogônia (8,77±1,14 e 9,04±0,96), espermatócitos (4,99±2,00 e 6,20±2,07; P<0,05), Leydig (8,05±2,82 e 7,89±2,29) e Sertoli (8,07±1,88 e 7,61±2,16; P<0,05). Houve correlação (P<0,05) entre os lados para o número de NORs: espermatócitos x Leydig (0,49); espermatócitos x Sertoli (0,49) e Leydig x Sertoli (0,96). Conclui-se ser válido o emprego da técnica AgNOR para avaliar o potencial proliferativo das células saudáveis em testículos de ovinos com prática execução e baixo custo.(AU)


The silver staining technique for AgNOR nucleolar organizer regions (NORs) is characterized by marking proteins linked to the ribosomal ribonucleic acid, evaluating cell proliferation. The aim was to study the validity of the AgNOR staining technique in the testicular cell proliferation in crossbred ovine. Forty-three pairs of ovine testicles between 6 and 10 months old were collected. Wilcoxon and Spearman tests were used with a significance level of 5%. The mean NORs count in cells of the right and left gonads were respectively: spermatogonia (8.77±1.14 and 9.04±0.96), spermatocytes (4.99±2.00 and 6.20±2.07, P<0.05), Leydig (8.05±2.82 and 7.89±2.29) and Sertoli cells (8.07±1.88 and 7.61±2.16; P<0.05). There was a correlation between the mean values for the right and left sides for the number of NORs (P<0.05) between Leydig x spermatocytes (0.49); spermatocytes x Sertoli (0.49) and Sertoli x Leydig (0.96). The study demonstrates that the AgNOR staining technique is indicated to evaluate the cell proliferative potential in ovine testis with practical implementation and low cost.(AU)

Objective: To study the chemical constituents of small polar in Aquilariae Lignum Resinatum. Methods: The column chromatography on silica gel, Sephadex LH-20, and semi-preparative HPLC was used to separate and purify the compounds, their structures were elucidated by NMR and MS spectroscopic data analyses. Results: Nine compounds were isolated from the petroleum ether of Aquilariae Lignum Resinatum and identified as 18-nor-abieta-8,11,13-triene-4α,7α-diol (1), 18-norabieta-8,11,13-triene-4α, 7β-diol (2), 7α-hydroxydehydroabietic acid (3), 2α-hydroxydehydroabietic acid (4), 2β-hydroxydehydroabietic acid (5), 7α,15- dihydroxy-methyl-dehydroabietate (6), 7-oxo-13β-hydroxyabiet-8(14)-en-18-oic acid (7), 2β-hydroxypimaric acid (8), and 3β- hydroxypimarol (9). Conclusion: All the compounds are isolated from the plants of Aquilaria Lam. for the first time, among them, compounds 1 and 2 are norditerpenoids and compounds 3-9 are diterpenoids.

ABSTRACT

Kerodon acrobata is a caviidae rodent endemic from Brazilian Cerrado. It was described only in 1997 and the data about it is very scarce. The aim of this work was to characterize the karyotype of K. acrobata. Giemsa staining, nucleolar organizer region (NOR) banding, C-positive heterochromatin banding and DAPI fluorescence were used in N metaphases of a specimen collected in Asa Branca Farm, in Aurora do Tocantins municipality, Tocantins state, Brazil. K. acrobata showed the same diploid number, fundamental number and chromosome morphology as Kerodon rupestris. But its NOR location and heterochromatin distribution patterns indicated a unique cytogenetic profile when compared to its sister species, emphasizing the evolutionary uniqueness of this relatively new and unknown species. This record also extends the distribution of this species northward.


Kerodon acrobata é um roedor caviídeo endêmico do Cerrado brasileiro. A espécie foi descrita apenas em 1997, e as informações sobre ela são muito escassas. O objetivo deste trabalho foi caracterizar o cariótipo K. acrobata. Coloração em Giemsa, bandeamento da região organizadora do nucléolo, bandeamento da heterocromatina C-positiva e fluorescência DAPI foram utilizados em N metáfases de um espécime coletado na fazenda Asa Branca, na cidade de Aurora do Tocantins, estado do Tocantins, Brasil. K. acrobata apresentou o mesmo número diplóide, número fundamental e morfologia dos cromossomos de Kerodon rupestris. Mas a localização de sua NOR e os padrões de distribuição de heterocromatina indicam um perfil citogenético único quando comparado com sua espécie irmã, enfatizando a singularidade evolutiva desta espécie pouco conhecida. Este registro também estende a distribuição desta espécie em direção ao norte.

ABSTRACT

This study described the karyotype of Geophagus cf. proximus. Specimens were collected in Água Preta Lake, Parque Ambiental de Belém, Pará State, Brazil. The karyotype were 2n = 48 chromosomes (FN = 60: 12M/SM+36ST/A) and no sexual chromosome differentiation. C-banding showed centromeric staining in all chromosomes. The first chromosome pair, besides centromeric coloration, presented a totally heterochromatic long arm. The nucleolus organizer regions (NORs) were studied by means of AgNO3. NORs were located at the short arm of the second chromosome pair.


Este estudo descreve o cariótipo de Geophagus cf. proximus. Espécimes foram coletados no lago Água Preta, Parque Ambiental de Belém, Estado do Pará, Brasil. O cariótipo obtido apresentou 2n = 48 cromossomos (NF = 60: 12M/SM+36ST/A), sem diferenciação de cromossomos sexuais. O bandeamento C mostrou marcações centroméricas em todos os cromossomos. O primeiro par cromossômico, além da coloração centromérica, apresentou o braço longo totalmente heterocromático. As regiões organizadoras do nucléolo (RONs) foram estudadas por meio da coloração de AgNO3. As RONs foram encontradas no braço curto do segundo par de cromossomos.

ABSTRACT

Increased fat intake is known to be a major cause of prostate cancer. In this study, we investigated the effect of dietary high fat on prostate intraepithelial neoplasia using transgenic adenocarcinoma mouse prostate (TRAMP) mice. Six-week-old male TRAMP mice were fed AIN93G (control group, 4.0 kcal/kg, n=6) and AIN93G-HFD (experimental group, 4.8 kcal/kg, n=7) for 10 weeks. Prostate histopathology, urogenital tract (UGT) weight, epididymal white adipose tissue weight, argyrophilic nucleolar organizer regions (AgNORs) counts, and serum leptin levels were examined. AIN93G-HFD fed group showed progressed neoplastic lesions in the prostate (P<0.05) compared to AIN93G fed group. AIN93G-HFD intake resulted in a increase in the weight of UGT (P<0.05) and epididymal white adipose tissue. The number of Ag-NOR positive dots significantly increased in each prostate lobe and final serum leptin levels in AIN93G-HFD fed group were about twice those of AIN93G fed group (P<0.05). Dietary high fat was related to the prostate cancer progression in the early stage of TRAMP mice and increased serum leptin levels, suggesting that the regulation of dietary components could delay the progression of prostate cancer.

ABSTRACT

Cytogenetic analyses were performed in four species of the Hypostominae subfamily, three from Hypostomus (Hypostomini) genus and Rhinelepis aspera (Rhinelepini). Three populations of Hypostomus ancistroides were analyzed, which had 2n=68 chromosomes, but presented different karyotype formulas. Hypostomus regani and H. strigaticeps, both from Ivaí river, showed 2n=72 chromosomes with two distinct cytotypes. In turn, R. aspera of the upper Paraná river basin presented 2n=54 chromosome. Multiple Nucleolar Organizer Regions (NORs) have been evidenced by silver nitrate staining in species of Hypostomus and single NOR in R. aspera. The observed variation in the chromosome number and the marked variability in karyotype formulas and NORs reveal a certain amount of karyotype variation in the genus Hypostomus suggesting the probable existence of cryptic species with independent chromosome traits. Therefore, our data can be of great value in discriminating species and understanding their chromosomal evolution.


Foram analisadas três populações de peixes identificadas como Hypostomus ancistroides, as quais apresentaram 2n=68 cromossomos, com distintas fórmulas cariotípicas. Hypostomus regani e H. strigaticeps, ambas do rio Ivaí, apresentaram 2n=72 cromossomos com citótipos distintos. Rinelepis aspera da bacia do alto rio Paraná apresentou 2n=54 cromossomos. Nucleolar Organizer Regions (NORs) múltiplas foram evidenciadas por nitrato de Prata para as espécies do gênero Hypostomus e NOR simples para R. aspera. A variação de número cromossômico observada, como também a acentuada variação nas fórmulas cariotípicas e nas NORs, são discutidas, sugerindo a existência de possíveis espécies crípticas com caracteres cromossômicos independentes. Portanto, nossos dados podem ser de grande valia na discriminação das espécies e no entendimento de sua evolução cromossômica.

ABSTRACT

Venezuelan scorpion envenomation is a public health problem produced by Tityus discrepans (TD) and Tityus zulianus (TZ) species. Patients-envenomend by TD developed gastrointestinal and pacreatic disorders and scorpion accidents involving TZ are associated with high mortality rate, which showed cardiopulmonary clinical disorders may be associated to the high levels of plasma catecholamines levels. This distinctive clinical output seems to be associated to a toxin repertoire diversity, which has been previously demonstrated. Trying to mimic the human-envenomation, some toxinological studies have been performed using TD and TZ venoms in several biomedels such as mice and anesthetized rams. The purpose of this study was to evaluate, in vivo using biomodels (mice), the role of autonomic nervous system (sympathetic) stimulation producing some of the clinical signs, via the catecholamines release, on the patho-physiology of the TZ and TD induced envenomation. Thus, a clinical signs here reported during a period of 1 hr, after a single intra-peritoneal injection of sub-lethal doses of TZ or TD venom, which are related with diarrhea, diaphoresis, intense salivation, dehydratation, dyspnea and spasticity in hind limbs. However, these animals did not exhibit vomiting, which is the most frequent human-envenomed TD patients. All animals inoculated with (TD or TZ) venoms develped diarrhea being more pronounced in TD group. Diaphoresis, sialorrhea and dehydratation were mainly observed in TD group. Dyspnea and the hind limb spasticity were only developed in TZ mice. These clinical manifestations (diarrhea, sialorrhea, dehydratation and intense salivation) are related to an activation of autonomic nervous system, via an intense release of their related neurotransmitters. Thus, autonomic stimulation (sympathetic) was evaluated following the catecholamine (Nor-Epinephrine) (NE) plasma levels in a function of envenomation time. El escorpionismo en Venezuela es un problema actual de salud pública producido por las especies de Tityus discrepans (TD) y Tityus zulianus (TZ). Los pacientes que presentan escorpionismo producido por TD desarrollan trastornos gastrointestinales y pancreáticos mientras que los afectados por TZ presentan una alta mortalidad y muestran una sintomatología relacionada a desordenes cardiopulmonares, los cuales parecen estar asociados a niveles elevados de las catecolaminas plasmáticas. Esta clínica diferente parece estar asociada a una composición distinta de toxinas de dichos venenos, lo cual ha sido previamente demostrado. En un intento de mimetizar o reproducir el escorpionismo en humanos se han realizado estudios toxinológicos con los venenos de TZ y TD utilizando varios biomodelos como son ratones y carneros anestesiados. El propósito de este trabajo fue evaluar, “in vivo” usando un Biomodelo (ratones), el papel de la estimulación del sistema nervioso autónomo (simpático) para producir algunos signos clínicos, vía la liberación de catecolaminas, en la fisiopatología del escorpionismo producido por TZ y TD. Así, los signos clínicos aquí descritos y observados durante 1 hr., después de la inyección de una dosis sub-letal de los venenos de TZ y TD, fueron la presencia de diarrea, diaforesis, salivación intensa, deshidratación, disnea y parálisis en las extremidades posteriores. Sin embargo, estos animales no presentaron vómitos, el cual es uno de los signos más frecuentemente observado en los pacientes con accidentes escorpiónicos por TD. Todos los animales inyectados con los venenos de TD y TZ presentaron diarrea especialmente en grupo TD. ABSTRACT

The active metabolite of vitamin D, 1,25-dihydroxyvitamin D3 (calcitriol), inhibits the growth of several types of human cancer cells in vitro, but its therapeutic use is limited because it causes hypercalcemia. Among its analogs, 19-nor-1,25-dihydroxyvitamin D2 (paricalcitol), has fewer calcemic effects and exhibits an activity equipotent to that of calcitriol. We assessed the antitumor and anti-inflammatory effects of paricalcitol in gastric cancer cells, and evaluated the potential role of vitamin D in the treatment of peritoneal metastatic gastric cancer. In this study, treatment with paricalcitol inhibited gastric cancer cell growth and induced cell cycle arrest. Paricalcitol also induced apoptosis and showed anti-inflammatory activity. Moreover, the growth of intraperitoneal metastases in vivo was reduced in mice treated with paricalcitol. 18F-FDG uptake was significantly lower in the paricalcitol group compared to control group (SUV; control group 13.2 +/- 5.3 vs paricalcitol group 4.5 +/- 3.0). Intraperitoneal tumor volume was significantly lower in paricalcitol treated mice (control group 353.2 +/- 22.9 mm3 vs paricalcitol group 252.0 +/- 8.4 mm3). These results suggest that the vitamin D analog, paricalcitol, has anticancer activity on gastric cancer cells by regulation of the cell cycle, apoptosis, and inflammation.

ABSTRACT

Análises citogenéticas de Potamotrygon aff. motoro e P. falkneri identificaram a ocorrência de um sistema múltiplo de cromossomos sexuais do tipo X1X1X2X2/X1X2Y, em ambas as espécies, com 2n = 66 cromossomos em fêmeas e 2n = 65 cromossomos nos machos. As regiões organizadoras de nucléolos (RONs) identificadas pela reação Ag-RON, evidenciaram marcações múltiplas em ambas as espécies (com variações de 5 a 7 RONs). A técnica de bandamento C, revelou a presença de blocos heterocromáticos localizados nas regiões centromérica em quase todos os cromossomos nas duas espécies em estudo. Através do presente estudo foi evidenciada uma heterogeneidade nos cariótipos, permitindo sugerir que rearranjos cromossômicos, como inversões e/ou translocações, ocorreram durante a evolução cromossômica nas duas espécies desse gênero.


Cytogenetic analysis of Potamotrygon aff. motoro and P. falkneri indicated the occurrence of an X1X1X2X2/X1X2Y multiple sex chromosome system in both species, with 2n = 66 chromosomes for females and 2n = 65 chromosomes for males. The nucleolus organizer regions (NORs) identified using Ag-NOR technique showed that both species have multiple Ag-NORs (5 to 7 chromosomes stained). C-banding technique indicated the presence of heterochromatic blocks in the centromeric regions of almost all chromosomes in both species. Through this study there was evidence of heterogeneity in the karyotypes, which suggests that chromosomal rearrangements such as inversions and/or translocations occurred during the chromosomal evolution in two species of this genus.

ABSTRACT

A 34-year-old male patient with suspected pheochromocytoma was referred to our department for paroxysmal hypertension. Pheochromocytoma was confirmed by very high plasma nor-metanephrine ( NMN ) and metanephrine ( MN ). Contrast CT of the adrenal disclosed a 3.0 cm x 3.0 cm mass in the left adrenal. 18 F-FDG-PET-CT showed high uptake only in the left adrenal. When the patient's blood pressure was well controlled with 4 mg/d doxazosin for 2 weeks, surgery was then performed under laparoscopy. A round solid left adrenal pheochromocytoma was resected. After surgery, plasma MN and NMN levels returned to normal, and the patient was free of clinical symptoms with normal blood pressure. This patient has been followed 3 years with normal blood pressure, MN, and NMN levels, without tumor recurrence shown hy adrenal CT.
